BioBrick_BASIC_DAHP Synthase (Aro4) (K229L)
Biology
This is the CDS for the yeast gene ARO4 which codes for the aromatic amino acid biosynthesis enzyme 3-deoxy-D-arabino-heptulosonate-7-phosphate (DAHP) synthase (E.C.2.5.1.54). An amino acid substitution K229L has been made which has been shown in the literature to lead to higher activity through deleting negative regulation. This in turn leads to higher rates of aromatic amino acid biosynthesis.
Usage
This part is immediately surrounded by BASIC integrated prefix and suffix (iP/iS), allowing the part to be used in BASIC assembly. Surrounding the BASIC iP/iS are the BioBrick prefix and suffix. This means that the part allows for cloning with both the BASIC assembly method and with BioBrick assembly. This part would naturally suit any projects looking to optimise or increase aromatic amino acid biosynthesis in Saccharomyces cerevisiae.
Characterisation
As a result of the Covid pandemic, we were unable to test this part in the lab, and so we can not currently confirm it functions as intended.
